Research Focus

Novel molecular therapeutics like siRNA, microRNAs or antagomiRs require potent extracellular and intracellular delivery devices to be effective antitumor agents.

For this purpose we design programmed nanoagents ( which recognize tumor cells by specific targeting mechanisms, invade the cells and release the bioactive therapeutic nucleic acid in the cytosol of target cells.

Potential Project:

Receptor-targeted, nucleic acid loaded nanoagents based on precise synthetic carrier polymers (see Schaffert, Angewandte Chemistry 2011; Nie, Biomaterials 2011) will be tested in tumor cell cultures for manipulation of cancer cells in a specific fashion (see for example Schaffert Pharm. Research 2011). Folate or tyrosine kinase receptor ligands will be applied for targeting, siRNAs and miRNAs manipulating the cell-cycle or phenotype of tumors will be applied.

Primary Techniques: Nanoparticle formation and characterization, delivery into cultured tumor cells, analysis of gene expression patterns, RT-QPCR, fluorescence microscopy, immunohistochemistry, flow cytometry, cell cycle analysis.
